Description

The NucleoBond™ AXG buffer sets include all the required buffers and enzymes (proteinase K and RNase A) for the extraction of genomic DNA from tissue and bacteria. Using these buffer sets in combination with NucleoBond™ AXG columns allows isolation of up to 500μg genomic DNA from different matrices. The quality of the genomic DNA is extraordinarily high and is suitable for all kinds of subsequent reactions. NucleoBond™ AXG columns run by gravity flow resulting in a high molecular weight DNA without shearing. Samples are first lysed with proteinase K. Before loading the lysate to the NucleoBond™ AXG column, it is diluted with buffer N2 in order to guarantee a good flow rate as well as a quantitative binding of the DNA. After efficient washing the genomic DNA is eluted with high-salt buffer and precipitated with isopropanol. NucleoBond™ AXG columns are available in three sizes (AXG 20, AXG 100 and AXG 500). NucleoBond™ buffer sets:NucleoBond™ buffer set III for the isolation of genomic DNA from bacteria: for 10 AXG 500 columns (maxipreps), 20 AXG 100 columns (midipreps) or 100 AXG 20 columns (minipreps). NucleoBond™ buffer set IV for the isolation of genomic DNA from tissue: for 10 AXG 500 columns (maxipreps), 20 AXG 100 columns (midipreps) or 100 AXG 20 columns (minipreps).
